This report describes the production and certification of a reference material of purified genomic DNA (gDNA) of Listeria monocytogenes (strain 4B, NCTC 11994). This CRM (IRMM-447) supports the harmonisation and validation of PCR methods by their use as taxonomic controls in PCR reactions. The homogeneity and stability (at four different temperatures for up to 8 weeks) of the batch was assessed by monitoring amplification of the prfA gene by PCR, by agarose gel electrophoresis of gDNA and determination of the amount of gDNA per vial. The material is stable at -20° C. The authenticity of the gDNA was confirmed by DNA sequence analysis of the prfA gene. Each vial contains an indicative mass of 1.1 µg freeze-dried gDNA, with an expanded uncertainty (Ucrm) of 0.7 µg (k = 2).
