Integrated microfluidic system that can: - Generate and manipulate (inject, merge) pL-size monodisperse emulsion droplets of water in oil for growing and manipulating microorganisms - Generate pL-size hollow capsules with the possibility of exchange with the environment via diffusion of growth substrate and effector molecules for growing and manipulating microorganisms - Screen and sort the generated emulsion droplets and capsules based on a fluorescence activated droplet sorting; FADS) One stand-alone microfluidic device must be purchased for a microfluidic droplet platform that allows microbial cells to be cultured in monodisperse emulsion droplets (pL volume) of aqueous growth medium in an oil phase where the encapsulated cells have limited exchange of molecules (other than gas exchange) with the surrounding droplets. The equipment must include a high-speed microscope camera that allows image analysis of the generated droplets. The equipment shall be capable of manipulating generated droplets so that fluorescent reagents, probes, or cells can be introduced by drop fusion and pico-injection. The device shall also allow the encapsulation of microbial cells in hollow capsules which allow the exchange of molecules and growth medium with the environment and which allow centrifugation and subsequent culture in new growth medium. It shall be possible to control the equipment remotely via e.g. WiFi so that it can be operated in a closed cabinet, e.g. anaerobic chamber. One additional stand-alone microfluidic device shall be purchased, which allows for high-throughput screening and sorting of microorganisms in emulsion droplets (water in oil), as well as hollow capsules containing microbial cells, based on fluorescence measurements from the emulsion droplets/capsules (FADS). The equipment must have an integrated high-speed microscope camera, as well as allow for sorting on fluorescence measurements with at least three different wavelengths in order to be able to multiplex different signals.
