Uni-HD.2024.692_Hybrid tandem mass spectrometer_ZMBH
A high-resolution hybrid tandem mass spectrometer in combination with an ultra-high-pressure liquid chromatography (UHPLC) system (LC-MS System) for highly sensitive LC-MS workflows will be procured. The proposed applications of the system include proteome analysis, identification of crosslinked peptides with amino acid resolution, and analysis of posttranslational modifications. The LC-MS system to be procured shall include the following components: • A mass spectrometer that combines quadrupole, linear ion trap and orbitrap mass analyzers and provides CID and HCD fragmentation techniques that can be applied at any MSn analysis stage. • An ion source with vertical adjustment options for optimizing spray conditions. • An ion source that generates internal calibrations for mass calibration in real time. • An ETD module that generates fluoranthene anions for electron transfer dissociation. • Installation of the LC-MS system and preparation for operation, including a quality check. • A UHPLC system for high-sensitivity LCMS workflows that supports both direct and trap-and-elute injections for low-flow applications in the range of 0.001-100 μL/min and is equipped with a column compartment and valve. • Specific parameters of the mass spectrometer: • The MS system shall provide a resolution of 500,000 FWHM at m/z 200. • The MS system shall have a quantitative proteomics workflow based on TMT marking technology with MSn functionality with high resolution and high mass accuracy (HRAM MSn). • The MS system must detect ions in real time and process the collected data so that specific peptides for MS3 fragmentation can be selected by real-time database searches during the measurement. • The MS system shall provide MSn functionality.
