CZ-Brno: chromatographs
The laboratory equipment required is used for the preparation, purification and characterisation of organic and biological substances. These are: polarimeter, point, ozonator, nitrogen monoxide analyser, electron paramagnetic resonance spectrometer and liquid chromatography assembly. The polarimeter will be used to analyse and characterise optically active organic compounds – intermediates and final products; the optical activity shall be determined with an accuracy of at least 0.005 degrees. The point shall be used to determine the melting point of organic compounds; i.e. intermediates and final products of planned organic synthesis; make it possible to determine accurately the melting point of three samples at the same time (three intermediates or products or one or two samples may contain calibration standards). Ozonator (laboratory source of ozone) will be used in organic synthesis of compounds with targeted biological activity; in particular, to carry out ozonolysis, which is a standard method by which chemoses selective splitting of the carbon-carbon double link can be carried out and relevant products containing the carbonyl group, i.e. aldehydes or ketones, can be generated. The Nitric Oxide analyser shall allow determination of the concentration of nitrogen oxide in the exhaled air in small animals and volunteers by means of online or off-line detection if this is not possible under the study conditions and the determination of nitrites, nitrates and S nitrosthiols in biological fluids and extracts of cell cultures and animal organs. Electron paramagnetic resonance spectrometer will be used to determine biologically relevant free radicals such as nitric oxide and various active forms of oxygen in biological samples (lyzates and extracts), as well as to determine these radicals in animal cells and tissues under physiological conditions. The liquid chromatography assembly will be used to determine the size and the exact molecular weight of the proteins, to identify aggregates in enzyme samples depending on the composition of the solution, to analyse oligomeric protein conditions in relation to pH and solution composition, and to analyse internal viscosity and protein compliance changes depending on ambient conditions. The minimum technical requirements are set out in the Annex to the specifications.
